This Technical Specification describes the measurement of moulds in ambient air in order to identify, quantify and characterize bioaerosol pollution in ambient air resulting from emissions from different sources. The method described specifies the sampling of moulds as part of the suspended particulate matter (SPM, here particles with aerodynamic diameter up to approximately 30 µm) using a filter sampling system with a gelatine/polycarbonate filter combination. The sampling duration can be varied between 10 min and 24 h. With the analytical method described here, the mesophilic and thermotolerant moulds are quantified by cultivation of the viable and culturable propagules on selective DG-18 agar. The Technical Specification DIN SPEC 91221 is a process standard and particularly deals with aspects of environmental protection focusing on microbiotic air pollution. It is directed at users from regulatory authorities, test institutes, research institutes as well as plant operators and equipment manufacturers. The Technical Specification DIN SPEC 91221 has been prepared by CEN/TC 264 "Air quality" with the intensive collaboration of German experts in WG 28 "Measurement of bioaerosols in ambient air and emissions") (secretariat and chairmanship: DIN).
